Mammary Tissue Digestion

Mammary tissue digestion is a laboratory process that separates mammary gland tissue into viable cellular components for biological analysis and culture. Enzymes such as collagenase and dispase break down extracellular matrix proteins, while gentle mechanical disruption, filtration, and centrifugation help release and enrich epithelial, stromal, and other mammary cell populations. The resulting cell suspensions support primary cell culture, organoid generation, flow cytometry, and studies of mammary development, lactation, and breast cancer. Careful control of enzyme exposure, temperature, and handling is essential to preserve cell viability, surface markers, and functional characteristics for reliable downstream experiments.

This protocol describes methods for purifying, quantitating, and characterizing extracellular vesicles (EVs)/exosomes from non-adherent/mesenchymal mammary epithelial cells and for using them to transfer mammary gland-forming ability to luminal mammary epithelial cells. EVs/exosomes derived from stem-like mammary epithelial cells can transfer this cell property to cells that ingest the EVs/exosomes.
