Midgut Dissection

Midgut dissection is a laboratory technique that isolates the midgut, the digestive tract region where organisms digest and absorb nutrients, for direct biological study. During the procedure, researchers carefully remove surrounding tissues, open or separate the gut in physiological saline, and preserve its structure for microscopy, staining, molecular analysis, or physiological assays. In biology, midgut dissection helps reveal epithelial organization, digestive function, nutrient transport, host–microbe interactions, and responses to toxins or pathogens. The method provides access to tissue-level processes that are difficult to observe in intact animals, supporting research in development, physiology, disease biology, and experimental genetics.

Microbial communities in mosquitoes hold great promise for vector biocontrol strategies. Most symbionts are uncultivable, requiring metagenomic analyses. We describe a method to dissect female mosquitoes and separate ovaries, midgut, and salivary glands preventing cross-contamination, facilitating microbiome studies at the organ level, and enhancing understanding of microorganisms' roles in mosquito biology.
