Cell Dispersion

Cell dispersion is the process of separating cells from one another or from a tissue matrix to produce a uniform suspension or distribute cells throughout a sample. It typically combines mechanical disruption with enzymatic dissociation, using agents such as trypsin or collagenase to break cell adhesion proteins and extracellular-matrix components while limiting damage to cell membranes. In biology, dispersed cells support primary cell culture, flow cytometry, microscopy, viability assays, and single-cell analysis. Careful control of enzyme exposure, temperature, and handling helps preserve cell survival and function, enabling reliable studies of cell behavior, tissue organization, disease mechanisms, and responses to experimental treatments.

Somitogenesis is a rhythmic developmental process that spatially patterns the body axis of vertebrate embryos. Previously, we developed transgenic zebrafish lines that use fluorescent reporters to observe the cyclic genes that drive this process. Here, we culture dispersed cells from these lines and image their oscillations over time in vitro.

Adapting Taylor Dispersion to Measure the Dispersion Coefficient of Electrolyte Solutions via an Accessible Microfluidic Setup

Here, we present a protocol to adapt the Taylor dispersion experiment to the microscale using microchannels fabricated in-house with a desktop craft cutter. The experimental platform can be used to compute the diffusion coefficient of single-species passive tracers and to visualize multispecies ion interaction and separation.

Detection of Biofilm Disassembly Through a Dispersion Assay

The video showcases a biofilm dispersion assay using a multi-well plate. Buffer and glucose treatment disassemble the biofilm, releasing bacteria, while bile salts induce stress, retaining bacteria in the protective extracellular polymeric substance. Higher dispersion with buffer and glucose confirms successful biofilm disassembly, estimated by colony forming units measurement.
