Mosquito Gut Colonization

Mosquito gut colonization is the establishment and persistence of microorganisms within a mosquito’s digestive tract, where microbial communities can influence host biology and pathogen transmission. After a blood meal or other feeding event, ingested microbes encounter the midgut, peritrophic matrix, digestive conditions, nutrient availability, and immune defenses that determine whether they are eliminated, persist, or interact with resident bacteria. Studying these processes helps explain mosquito microbiome development and vector competence, including how gut microbes affect the survival and replication of pathogens such as malaria parasites and arboviruses. This knowledge supports research on microbial-based strategies to reduce disease transmission.

Source: Liu, X., et al. A Bacterial Oral Feeding Assay with Antibiotic-Treated Mosquitoes. J. Vis. Exp. (2020)This video demonstrates the depletion of native gut microbiota in mosquitoes using an antibiotic-containing sucrose solution. This is followed by controlled recolonization with a bacterial suspension delivered through a membrane feeding system to establish a defined bacterial population in the mosquito gut.

Depletion of Ribosomal RNA for Mosquito Gut Metagenomic RNA-seq

A ribosomal RNA (rRNA) depletion protocol was developed to enrich messenger RNA (mRNA) for RNA-seq of the mosquito gut metatranscriptome. Sample specific rRNA probes, which were used to remove rRNA via subtraction, were created from the mosquito and its gut microbes. Performance of the protocol can result in the removal of approximately 90-99% of rRNA.

Source: Liu, X., et al. A Bacterial Oral Feeding Assay with Antibiotic-Treated Mosquitoes. J. Vis. Exp. (2020)This video demonstrates the dissection of the mosquito midgut to isolate gut bacteria. The midgut is extracted from the digestive tract, homogenized to release the bacteria, and plated on nutrient agar to culture bacterial colonies.
