Oxygen-rich Environment

An oxygen-rich environment is a setting with oxygen concentrations or partial pressures above those of typical ambient air, influencing how organisms and cells generate energy and respond to stress. Greater oxygen availability can enhance diffusion into tissues and support aerobic respiration, in which mitochondria use oxygen as the terminal electron acceptor to produce ATP. However, elevated oxygen can also increase reactive oxygen species, prompting antioxidant defenses and changes in gene expression, metabolism, and cellular survival. In biology, oxygen-rich conditions are studied in cell culture, developmental research, environmental physiology, and disease models to examine metabolism, oxidative stress, tissue responses, and adaptation.

Here, we present a method of the spectroscopic characterization of organic molecules by means of time-resolved photoluminescence spectroscopy on the nanosecond-to-millisecond timescale in oxygen-free conditions. Methods to efficiently remove oxygen from the samples and, thus, limit luminescence quenching are also described.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
