Phenol Chloroform Extraction

Phenol-chloroform extraction is a liquid-liquid separation method used to purify nucleic acids from biological samples by removing proteins, lipids, and other cellular components. In an aqueous mixture, phenol denatures proteins while chloroform enhances separation into organic and aqueous phases; after centrifugation, nucleic acids generally remain in the aqueous layer, which can be recovered and precipitated with alcohol and salt. The method supports DNA and RNA isolation for molecular biology applications such as cloning, PCR, sequencing, and gene-expression analysis, although it requires careful handling because phenol and chloroform are hazardous and residual solvent can inhibit downstream reactions.
