Protein Localization

Protein localization is the study of where proteins are produced, transported, and positioned within cells, a distribution that enables them to perform specific biological functions. Cells direct proteins through targeting signals, such as amino acid sequences, that guide transport into organelles, across membranes, or to particular cellular regions; localization can also change in response to cellular conditions. Researchers use fluorescence microscopy, tagged proteins, and cell fractionation to map these destinations and examine how mislocalization affects cell behavior. Protein localization analysis supports research in cell biology, disease mechanisms, drug development, and the design of engineered proteins.

Identification of Plasmodesmal Localization Sequences in Proteins In Planta

Plant intercellular connections, the plasmodesmata (Pd), play central roles in plant physiology and plant-virus interactions. Critical to Pd transport are sorting signals that direct proteins to Pd. However, our knowledge about these sequences is still in its infancy. We describe a strategy to identify Pd localization signals in Pd-targeted proteins.

Nano-fEM: Protein Localization Using Photo-activated Localization Microscopy and Electron Microscopy

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.

This video demonstrates a method for monitoring infected cell protein 0, ICP0 trafficking in herpes simplex virus-1 infection. Post-de novo synthesis, ICP0 translocates to the nucleus, later moving to the cytoplasm during infection progression. Immunofluorescence microscopy reveals and analyzes the protein's subcellular localization, offering insights into its trafficking across infection phases.

An Immunolabeling Technique to Visualize the Subcellular Localization of Proteins

This video demonstrates a technique for the immunolabeling of sigma-1 receptors in a murine heart tissue section using quantum dots. The heart tissue is fixed, osmicated, stained, dehydrated, embedded in resin, and then sectioned. The sections are then treated with antibodies and labeled with quantum dots to demarcate the subcellular localization of Sigma-1 receptors using electron microscopy.
