Single Channel

Single channel analysis is the measurement of electrical activity produced by an individual ion channel, revealing how membrane proteins regulate the movement of ions across cell membranes. In a patch-clamp experiment, a glass micropipette seals onto a small membrane region, while changes in voltage or chemical conditions cause channels to open and close; the resulting picoampere currents are recorded as discrete events. These measurements identify conductance, opening and closing kinetics, and channel responses to ligands or drugs. Single-channel recordings help explain neuronal signaling, muscle excitation, sensory transduction, and disease-related channel dysfunction, while supporting the development of therapies that target ion-channel activity.

GABA-activated Single-channel and Tonic Currents in Rat Brain Slices

We use the patch-clamp technique to measure GABA-activated single-channel currents (GABAA channels, GABAA receptors) and the synaptic and tonic currents they generate in neurons. Activation of the channels decreases neuronal excitability in health and disease 1,2,3,4.

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

A Proteoliposome-Based Efflux Assay to Determine Single-molecule Properties of Cl- Channels and Transporters

Proteoliposomes are used to study purified channels and transporters reconstituted in a well-defined biochemical environment. An experimental procedure to measure efflux mediated by these proteins is illustrated. The steps to prepare proteoliposomes, perform the recordings, and analyze data to quantitatively determine the functional properties of the reconstituted protein are described.

This paper discusses how to build a brain-computer interface by relying on consumer-grade equipment and steady-state visually evoked potentials. For this, a single-channel electroencephalograph exploiting dry electrodes was integrated with augmented reality glasses for stimuli presentation and output data visualization. The final system was non-invasive, wearable, and portable.
