Stromal Thymic Interactions

Stromal thymic interactions are the coordinated contacts and signals between thymic stromal cells and developing T lymphocytes that establish a functional, self-tolerant T-cell repertoire. Thymic epithelial cells, fibroblasts, and related stromal populations provide structural niches and molecular cues, including Notch ligand DLL4, cytokines such as interleukin-7, and self-antigen presentation by medullary epithelial cells, guiding thymocyte survival, differentiation, positive selection, and negative selection. These interactions are central to immune system development and tolerance, and their disruption can impair T-cell production or promote autoimmunity. Studying them informs research on immunodeficiency, autoimmune disease, aging, and strategies for thymus regeneration.

Three-Dimensional Thymic Culture System to Generate iPSC-Derived Thymic Emigrants

2025

This video demonstrates a protocol for generating functional antigen-specific T cells from iPSC-derived immature T cells using a three-dimensional thymic culture system. Co-culturing the iPSC-derived immature T cells with endogenous lymphocyte-depleted mice fetal thymic lobes resulted in successful maturation of the T cells to form CD8αβ+ MHC class I+ iPSC-derived thymic emigrants (iTEs).

We describe the preparation of thymic slices that, in combination with flow cytometry, can be used to model positive and negative selection of developing T cells. Thymic slices can also be adapted for the in situ analysis of thymocyte migration, localization, and signaling via immunofluorescence and two-photon microscopy.

Here we present a protocol to co-culture in three-dimensions, which is useful for investigating multicellular interactions and extracellular matrix-dependent modulation of cancer cell behavior. In this experimental model, cancer cells are cultured on collagen gels embedded with human cancer-associated fibroblasts.

We describe methodologies for establishing in vitro heterotypic three-dimensional models comprising ovarian fibroblasts and normal ovarian surface or ovarian cancer epithelial cells. We discuss the use of these models to study stromal-epithelial interactions that occur during ovarian cancer development.
