Density Gradient Centrifugation

Density gradient centrifugation is a separation technique that resolves particles, organelles, cells, or macromolecules according to their size, shape, and density. During centrifugation, a sample moves through a medium whose density gradually changes, causing components to sediment at different rates or migrate until they reach a layer matching their buoyant density. In biology, this method helps isolate nuclei, mitochondria, lysosomes, viruses, DNA, and other cellular components for structural and functional analysis. By producing relatively pure fractions while preserving biological material, density gradient centrifugation supports cell biology, molecular research, diagnostics, and studies of cellular organization.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...
