Terminal Procedure

A terminal procedure is a biological research procedure that ends an experiment by humanely euthanizing an organism, often to obtain tissues or other endpoint measurements. It typically follows approved experimental endpoints and uses a species-appropriate method to minimize pain and distress, after which researchers may collect blood, organs, or tissue for histology, molecular assays, or biochemical analysis. In preclinical biology, terminal procedures enable measurements that cannot be made in living subjects, helping link treatment exposure to physiological, cellular, and pathological outcomes while supporting ethical study design, reproducibility, and regulatory oversight.

Loading Drosophila Nerve Terminals with Calcium Indicators

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.
