Tick Saliva Mirnas

Tick saliva microRNAs (miRNAs) are small, noncoding RNA molecules released during tick feeding that can regulate gene expression and shape interactions between ticks and their hosts. After entering host tissues, often within extracellular vesicles, these miRNAs can bind complementary sequences in target messenger RNAs, reducing translation or promoting transcript degradation; in the tick, they may also help control salivary-gland function and feeding-related processes. Studying these molecules clarifies how ticks modulate host immune and inflammatory responses while obtaining a blood meal. Their stability and host effects support research into biomarkers of tick exposure, anti-tick vaccines, and strategies to limit transmission of tick-borne pathogens.

The collection of infected tick hemolymph, salivary glands, and saliva is important to study how tick-borne pathogens cause disease. In this protocol we demonstrate how to collect hemolymph and salivary glands from feeding Ixodes scapularis nymphs. We also demonstrate saliva collection from female I. scapularis adults.

Here, we present a protocol to collect saliva, haemolymph, salivary glands, and midgut, following the dissection of an individual fed tick, to study tissue-specific localization of tick-borne pathogens to advance understanding of vector competence and pathogen-microbiome interactions.
