Whole Mount Preparation

Whole mount preparation is a biological technique that preserves and displays an entire small specimen or tissue rather than cutting it into sections, allowing researchers to examine its overall organization. The specimen is typically fixed to maintain structure, permeabilized or cleared to improve visibility, stained or labeled to reveal selected components, and mounted on a slide for microscopy. In biology, whole mounts help visualize anatomical patterns, embryonic development, organ structures, cell distributions, and tissue-specific markers in their spatial context. This approach complements histological sectioning by retaining three-dimensional relationships and providing a broader view of morphology and developmental processes.

The zebrafish embryo is an excellent model for developmental biology research. During embryogenesis, zebrafish develop with a yolk mass, which presents three-dimensional challenges for sample observation and analysis. This protocol describes how to create two-dimensional flat mount preparations of whole mount in situ (WISH) stained zebrafish embryo specimens.
