Automated Cell Counter

An automated cell counter is a laboratory instrument that rapidly measures the number and, in many systems, the concentration and viability of cells in a sample. It typically analyzes cells by detecting optical features in images or changes in electrical impedance as cells pass through a sensing region, while software distinguishes individual cells and calculates the results. In biology, automated cell counting supports cell culture maintenance, seeding-density adjustment, growth monitoring, and preparation of samples for assays. By reducing manual counting with a hemocytometer, these systems can improve throughput, consistency, and reproducibility, although accurate measurements still depend on appropriate sample preparation and instrument settings.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.

The goal of this protocol is to manufacture pathogen-specific clinical-grade T cells using a bench-top, automated, second generation cell enrichment device that incorporates a closed cytokine capture system and does not require dedicated staff or use of a GMP facility. The cytomegalovirus pp65-specific-T cells generated can be directly administered to patients.
