Yellow Fluorescent Protein

Yellow fluorescent protein (YFP) is a genetically encoded fluorescent reporter derived from green fluorescent protein that emits yellow light, making invisible biological events observable in living cells. After the protein folds, an internally formed chromophore absorbs blue or blue-green light and releases lower-energy yellow fluorescence, while mutations tune its spectral properties and can alter sensitivity to pH or halide ions. Researchers attach YFP to proteins, promoters, or organelles to track gene expression, protein localization, and cellular dynamics using fluorescence microscopy. YFP can also serve as a Förster resonance energy transfer acceptor, enabling studies of molecular interactions, signaling pathways, and physiological changes in real time.

We describe a comprehensive and practical protocol for fluorescence recovery after photobleaching experiments with live cells. Although the protocol was used to measure the mobility of yellow fluorescent protein-tagged p62 in aggresome-like induced structures, it can be applied to a variety of microscopy systems and fluorescent proteins.
