Immunocompromised Nude Mouse

An immunocompromised nude mouse is a laboratory mouse with a genetic defect that causes an absent or underdeveloped thymus and greatly reduces functional T-cell immunity, making it valuable for cancer research. Because these mice have limited cellular immune responses, human tumor cells or tissue can be implanted as xenografts and studied without rapid rejection; innate immune components remain available to influence tumor growth and treatment response. Researchers use nude mouse models to investigate tumor development, metastasis, tumor-host interactions, and the effectiveness of anticancer drugs, radiation, and targeted therapies. These models help connect laboratory findings with preclinical cancer studies while highlighting the role of immunity in disease progression.

Mycobacterium leprae, the causative agent of leprosy, does not grow in vitro. We describe an easy to follow protocol to prepare a bacillary suspension to ensure the maintenance of large quantities of M. leprae for a variety of applications. Protocols for propagation by mouse footpad inoculation, evaluation of viability, freezing and thawing bacillary stock are described in detail.
