Macrophage Engraftment

Macrophage engraftment is the establishment and persistence of transferred or engineered macrophages within a tissue, providing a way to study and manipulate immune responses. After administration, macrophages migrate into the target microenvironment, interact with local cells and extracellular matrix, and may survive, proliferate, or adopt tissue-specific functional states. In cancer research, this approach helps evaluate how macrophages influence tumor growth, immune suppression, metastasis, and responses to therapy. Engrafted macrophages can also serve as vehicles for studying targeted delivery, genetic engineering, and strategies that reprogram tumor-associated macrophages toward antitumor activity, supporting the development of cell-based and immunomodulatory treatments.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.

Tracking Dynamics of Muscle Engraftment in Small Animals by In Vivo Fluorescent Imaging

We describe an in vivo fluorescence imaging protocol to monitor muscle regeneration by GFP-labeled myoblasts after transplantation into skeletal muscles of both healthy and dystrophic mice. This protocol can be adapted to study muscle regeneration by transplantation of other types of cells and in other muscular conditions as well.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.
