Metastatic Outgrowth

Metastatic outgrowth is the expansion of disseminated cancer cells into clinically detectable secondary tumors after they colonize distant tissues, making it a critical stage in cancer progression and mortality. It occurs when surviving tumor cells adapt to a new tissue microenvironment, exit dormancy, proliferate, recruit blood vessels, and evade immune destruction. Studying this process helps researchers identify how tumor–stroma interactions, angiogenesis, and organ-specific niches support metastatic growth. Experimental models of metastatic outgrowth are used to evaluate disease progression, discover biomarkers of recurrence, and develop therapies that target disseminated cells or the environments that enable them to form secondary tumors.

Immunofluorescence Assay to Evaluate Effect of Target Proteins on Neurite Outgrowth

In this video, we demonstrate an EGFP-based immunofluorescence assay to identify the efficacy of target proteins on the neurite outgrowth of primary cortical neurons. The primary neurons are transfected with a fluorescent protein, EGFP, and a target protein, and then an immunoassay is performed to investigate the effect of the target protein on neurite outgrowth.

An In Vitro System to Study Tumor Dormancy and the Switch to Metastatic Growth

A modified 3-D in vitro system is presented in which growth characteristics of several tumor cell lines in reconstituted basement membrane correlate with the dormant or proliferative behavior of the tumor cells at a metastatic secondary site in vivo.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
