Neutrophil-lineage Analysis

Neutrophil-lineage analysis is the study of neutrophils and their developmental stages, identities, and functions in tissues, including tumors. It distinguishes lineage states by combining cellular morphology with surface-marker, gene-expression, or functional measurements, allowing researchers to track maturation and changes caused by local signals. In cancer research, this analysis helps define how neutrophil populations enter and adapt to the tumor microenvironment, where they may influence inflammation, immune suppression, angiogenesis, and metastasis. The resulting profiles support investigation of tumor biology, biomarker development, and assessment of therapies that alter myeloid-cell behavior.

This video demonstrates the induction and visualization of neutrophil extracellular traps or NETs in pre-labeled neutrophils using patient serum containing anti-neutrophil cytoplasmic antibodies. Furthermore, using an impermeable DNA dye and fluorescence microscopy enables the visualization of NETs.

Neutrophils are the most abundant type of white blood cells. The isolation of neutrophils from human blood by density gradient separation method and the differentiation of human promyelocytic (HL60) cells along the granulocytic pathway are described here; to test their role on sensitivity of lymphoma cells to anti-lymphoma agents.
