Nod/scid Mouse Preparation

NOD/scid mouse preparation is the controlled process of selecting, housing, handling, and conditioning non-obese diabetic/severe combined immunodeficient mice for cancer experiments, where their impaired adaptive immunity enables human tumor cells to engraft. These mice lack functional T and B lymphocytes, and the NOD genetic background further reduces innate immune activity, so aseptic technique, specialized housing, and careful monitoring are essential during preparation. Properly prepared animals support tumor xenograft establishment, assessment of tumor growth, and evaluation of therapeutic responses in vivo. This model helps researchers study human cancer biology and drug efficacy while standardizing variables that influence engraftment, animal welfare, and experimental reproducibility.

2019

This protocol provides a method to establish humanized mice (hu-NSG) via intrahepatic injection of human hematopoietic stem cells into radiation-conditioned neonatal NSG mice. The hu-NSG mouse is susceptible to HIV infection and combinatorial antiretroviral therapy (cART) and serves as a suitable pathophysiological model for HIV replication and latency investigations.

Imaging Transgenic Beta-Cells in an Immunodeficient Mouse Challenged with Diabetic Splenocytes

2025

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.

A Technique to Generate a Human PBMC-Engrafted Humanized Xenograft Mouse Model

2025

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.
