Organ-specific Delivery

Organ-specific delivery is a targeted drug-delivery strategy that directs therapeutic agents to a particular organ, improving treatment localization while limiting exposure to healthy tissues. In cancer research, engineered carriers or targeting molecules can exploit organ-specific blood flow, tissue barriers, molecular receptors, or tumor-associated features to concentrate a payload at the intended site and regulate its release. This approach may enhance anticancer efficacy, reduce systemic toxicity, and support delivery of chemotherapy, nucleic acids, proteins, or imaging agents. Researchers use organ-specific delivery to investigate tumor biology, improve treatment precision, and develop therapies tailored to cancers arising in distinct anatomical environments.

This manuscript describes an ex vivo model system comprised of organ-conditioned media derived from the lymph node, bone, lung, and brain of mice. This model system can be used to identify and study organ-derived soluble factors and their effects on the organ tropism and metastatic behavior of cancer cells.

Intubation-mediated intratracheal (IMIT) instillation of reagents is an excellent, noninvasive method for studying respiratory disease, as well as a method for instilling therapeutic reagents directly into the lung. It is a rapid and highly reproducible method which is suitable for preclinical testing.

Tissue-specific microRNA inhibition is a technology that is underdeveloped in the microRNA field. Herein, we describe a protocol to successfully inhibit the miR-181 microRNA family in myoblast cells from the heart. Nanovector technology is used to deliver a microRNA sponge that demonstrates significant in vivo cardio-specific miR-181 family inhibition.
