Pdx Model Establishment

Patient-derived xenograft (PDX) model establishment is a cancer research method that grows human tumor tissue in immunodeficient animals to preserve clinically relevant features of a patient’s cancer. The process typically involves collecting a fresh tumor specimen, implanting tissue fragments or cells into a host mouse, and monitoring engraftment and growth through serial transplantation. Once established, PDX models can retain aspects of tumor heterogeneity, histology, and treatment response, supporting preclinical evaluation of anticancer drugs and combination therapies. They also provide a platform for studying tumor biology and may help inform personalized treatment strategies, although their lack of a fully functional human immune system limits some applications.

A method is described to create organoids using patient-derived xenografts (PDX) for in vitro screening, resulting in matched pairs of in vivo/in vitro models. PDX tumors were harvested/processed into small pieces mechanically or enzymatically, followed by the Clevers’ method to grow tumor organoids that were passaged, cryopreserved and characterized against the original PDX.
