Tumor Metabolism

Tumor metabolism is the study of how cancer cells alter energy production and nutrient use to support growth, survival, and adaptation within tumors. In many tumors, oncogenic signaling and conditions such as hypoxia increase glucose uptake and glycolysis, producing lactate even when oxygen is available, while metabolic pathways redirect nutrients toward nucleotide, lipid, and protein synthesis. These changes interact with the tumor microenvironment and can promote invasion, treatment resistance, and immune evasion. In cancer research, metabolic profiling and studies of enzymes, metabolites, and nutrient dependencies help identify biomarkers and therapeutic targets, including strategies that disrupt glycolysis or tumor-specific metabolic vulnerabilities.

2025

Here, we establish a metabolic profiling platform integrating three-dimensional (3D) glioma spheroids and imaging-based normalization via extracellular flux analysis. This approach enables more accurate assessment of tumor metabolic responses to drug treatments and provides insights into potential mechanisms of resistance.

Extracellular Flux Assay in 3D Spheroids: A Method for Measuring the Metabolic Activity of Pancreatic Tumor Spheroids

2023

This protocol describes the technique for performing extracellular flux assay on 3D spheroids generated from the co-culture of pancreatic cancer cells (Patu8902) and activated pancreatic fibroblasts (PS1) to measure the two major energy-producing pathways, glycolysis and mitochondrial respiration.

FDG Based PET/CT Scan: A Method of Using Radiotracer for Non-invasive Imaging of Tumor Metabolism

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

What is Metabolism?

2019

Overview Metabolism represents all of the chemical activity in a cell, including reactions that build molecules (anabolism) and those that break molecules down (catabolism). Anabolic reactions require energy, whereas catabolic reactions provide it. Thus, metabolism describes how cells transform energy through a variety of chemical reactions, which are often made more efficient with the help of enzymes. Metabolism Is the Sum of All of the Chemical Reactions That Take Place in an Organism
