Surgical Microscope

A surgical microscope is an optical instrument that provides magnified, illuminated views of small anatomical structures, supporting precise observation and manipulation during medical procedures. Its objective lenses, binocular eyepieces, adjustable magnification, and focused lighting enlarge the operative field while preserving depth and detail. In cancer research, surgical microscopes help investigators examine tumor tissue, identify boundaries between malignant and healthy structures, and perform delicate tissue collection or microsurgical procedures. These capabilities improve specimen quality, support studies of tumor architecture and invasion, and contribute to research on image-guided surgery and more precise cancer treatment.

Keeping the microscope optics clean is important for high-quality imaging. Dust, fingerprints, excess immersion oil, or mounting medium on or in a microscope causes reduction in contrast and resolution. DIC is especially sensitive to contamination and scratches on the lens surfaces. This protocol details the procedure for keeping the microscope clean.

Major Components of the Light Microscope

2008

The light microscope is a basic tool for the cell biologist, who should have a thorough understanding of how it works, how it should be aligned for different applications, and how it should be maintained as required to obtain maximum image-forming capacity and resolution. The components of the microscope are described in detail here.

Microscopic Visualization of Bacterial Microlesions in Mouse Heart Tissue

Source: Brown, A. O., et al. Visualization of Streptococcus pneumoniae within Cardiac Microlesions and Subsequent Cardiac Remodeling. J. Vis. Exp. (2015).This video demonstrates the microscopic visualization of bacterial microlesions in mouse heart tissue. The heart of a mouse infected with Streptococcus pneumoniae is harvested, washed, and fixed before being processed to obtain coronal sections. These sections are stained with hematoxylin and eosin and examined under a microscope to visualize...

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Surgical Preparation and Physiological Monitoring This video provides an introductory overview of surgical preparation, intraoperative monitoring, and postoperative care in swine. It is intended to familiarize learners with key...

Microscopic Anatomy of Skeletal Muscles

2024

Skeletal muscle cells, also called muscle fibers, are distinctly elongated, multi-nucleated, slender biological units. They are packed with specialized structures designed to facilitate their primary function, which is contraction. The muscle sarcolemma is a plasma membrane enclosing each muscle cell that conducts electrical signals called action potentials. The sarcolemma extends into the cell to form T-tubules, ensuring the neural impulses are uniformly distributed across the entire muscle...
