Pancreatic Cancer Stem Cells

Pancreatic cancer stem cells are a subset of tumor cells with self-renewal and differentiation capacity, and they are studied because they may sustain tumor growth and contribute to disease recurrence. Their behavior depends on stem-like regulatory programs and interactions with the tumor microenvironment, allowing some cells to initiate tumors, adapt to stress, and survive treatment. Research in cancer biology uses pancreatic cancer stem cell models to investigate tumor initiation, metastasis, drug resistance, and cellular heterogeneity. Understanding these populations may support the development of therapies that target both rapidly dividing tumor cells and treatment-resistant cells that help drive long-term disease progression.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.
