Nanoparticle Signal Enhancement

Nanoparticle signal enhancement is the increase in a measurable analytical response produced by incorporating nanoscale materials into a chemical sensing or detection system. Nanoparticles can concentrate analytes at their surfaces, provide high surface area for molecular interactions, and, in plasmonic materials such as gold or silver, amplify electromagnetic fields through localized surface plasmon resonance; these effects strengthen optical or electrochemical signals. In chemistry, the approach supports more sensitive detection of trace compounds, biomolecules, and reaction products, including surface-enhanced Raman measurements and colorimetric assays. Its value lies in improving detection limits while enabling smaller sample volumes and more selective molecular analysis.

2025

Take cultures of human monocyte-derived microglia-like cells.Introduce nanoparticles containing an anti-inflammatory drug into the experimental culture and the free drug into the control. The nanoparticles feature a hydrophilic shell and a hydrophobic core encapsulating the lipophilic drug. The cells endocytose the nanoparticles, which release the drug intracellularly, enhancing its delivery. The free drug enters by passive diffusion, a process constrained by the concentration gradient, which...

An Assay to Screen Bioactive Nanoparticles for Toll-Like Receptor Signaling Inhibition

2025

This video demonstrates a reporter cell-based assay for screening potential bioactive nanoparticles that inhibit Toll-like receptor (TLR) signaling. The assay involves introducing lipopolysaccharides (LPS) mixed with peptide-gold nanoparticle hybrids to reporter macrophages expressing the reporter proteins — secreted embryonic alkaline phosphatase (SEAP) and luciferase. The reduction of LPS-induced TLR signal mediated by the hybrids is determined by measuring the reporter signals.
