Photon Interaction

Photon interaction is the way light particles exchange energy with matter, a process that underlies many chemical changes and analytical measurements. When a molecule absorbs a photon with suitable energy, it can move from its ground state to an excited electronic, vibrational, or rotational state; the excited species may then emit light, transfer energy, or undergo a photochemical reaction. In chemistry, these interactions support spectroscopy, fluorescence analysis, photochemistry, and the study of molecular structure and dynamics. Understanding photon energy, absorption wavelengths, and relaxation pathways helps researchers identify substances, control light-driven reactions, and develop materials and processes for energy, sensing, and environmental applications.

In vivo Quantification of G Protein Coupled Receptor Interactions using Spectrally Resolved Two-photon Microscopy

By employing a spectrally resolved two-photon microscopy imaging system, pixel-level maps of Förster Resonance Energy Transfer (FRET) efficiencies are obtained for cells expressing membrane receptors hypothesized to form homo-oligomeric complexes. From the FRET efficiency maps, we are able to estimate stoichiometric information about the oligomer complex under study.

We have developed novel laboratory tools and protocols for intravital imaging acquisition of the thymus. Our technique should help in the identification of “niches” within the thymus where T cell development occurs.
