Ultracentrifuge Operation

Ultracentrifuge operation is the controlled use of extremely high-speed rotation to separate particles, molecules, or colloids according to their sedimentation behavior, making it an important analytical and preparative method in chemistry. A sample is placed in a balanced rotor and spun under controlled conditions; centrifugal force drives components through the liquid at rates determined by mass, shape, buoyant density, and solvent viscosity. Chemists use differential and density-gradient centrifugation to fractionate mixtures, characterize macromolecules, isolate nanoparticles, and study aggregation or molecular interactions. Careful control of rotor speed, temperature, sample loading, and run time improves resolution, reproducibility, and instrument safety.
