Canonical Wnt Expression

Canonical Wnt expression refers to the spatial and temporal production of Wnt ligands and pathway components that regulate cell behavior during development. In the canonical pathway, Wnt binding to Frizzled and LRP5/6 receptors inhibits the β-catenin destruction complex, allowing β-catenin to accumulate, enter the nucleus, and activate TCF/LEF-dependent genes. In developmental biology, analyzing these expression patterns helps explain embryonic axis formation, tissue patterning, stem-cell maintenance, and cell-fate decisions. Expression mapping and pathway perturbation can reveal how local signaling gradients coordinate morphogenesis and how abnormal Wnt activity contributes to developmental defects or disease.

Non-Canonical Wnt Signaling Pathways

Wnt is a zygotic effect gene that is expressed during very early embryonic development. It regulates various processes in animals starting from early development through the adult stage, such as organogenesis in the embryo and maintenance of neuronal and blood stem cells. Wnt proteins can induce a wide variety of intracellular pathways depending upon the specific abilities of different Wnt ligands to form a complex with shared and cognate receptors in the presence of different co-receptors. The...

The gene encoding the main signaling molecules of the Wnt signaling pathways (the Wnt proteins) was discovered almost four decades ago by Nüsslein-Volhard and Wieschaus. They identified and originally named the gene "wingless" (wg) after a phenotype discovered during their landmark genetic screen in Drosophila for body pattern defects. At around the same time, another researcher named Harold Varmus found that a murine tumor virus activates the mammalian wg homolog, Int-1, which results in tumor...

The use of reporter mice coupled to whole mount and section staining, microscopy and in vivo assays facilitates the analysis of mechanisms underlying the normal patterning of the respiratory tract. Here we describe how these techniques contributed to the analysis of Wnt signaling during tracheal development.
