H2a.f/z-egfp

H2a.f/z-egfp is a fluorescent chromatin reporter that labels cell nuclei by combining the histone H2A.F/Z with enhanced green fluorescent protein (EGFP). After expression, the H2A.F/Z-EGFP fusion protein is incorporated into nucleosomes, coupling EGFP fluorescence to chromatin and enabling visualization of nuclear structure and behavior in living cells. In developmental biology, this reporter supports real-time imaging of nuclear movements, cell division, tissue organization, and cellular dynamics as embryos and developing tissues change. Its signal can help researchers follow developmental events without relying solely on fixed samples, linking chromatin-associated fluorescence with the spatial and temporal progression of development.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.

In Parkinson's Disease (PD), Substantia Nigra (SNc) dopaminergic neurons degenerate, leading to motor dysfunction. Here we report a protocol for culturing ventral midbrain neurons from a mouse expressing eGFP driven by a Tyrosine Hydroxylase (TH) promoter sequence, harvesting individual fluorescent neurons from the cultures, and measuring their transcriptome using RNA-seq.
