Inner Ear Immunostaining

Inner ear immunostaining is a laboratory method that uses labeled antibodies to identify and visualize specific proteins, cells, or structures within developing inner-ear tissue. The technique relies on antibody-antigen binding: a primary antibody recognizes a target molecule, while a fluorescently or enzymatically labeled detection system reveals its location in fixed tissue sections or whole-mount preparations. In developmental biology, inner ear immunostaining helps map the emergence and organization of sensory hair cells, supporting cells, neurons, and other tissue components over time. These spatial and temporal patterns provide evidence about cell differentiation, tissue organization, developmental signaling, and changes associated with genetic or experimental perturbations.

Source: Richard Glickman-Simon, MD, Assistant Professor, Department of Public Health and Community Medicine, Tufts University School of Medicine, MA This video describes the examination of the ear, beginning with a review of its surface and interior anatomy (Figure 1). The cartilaginous auricle consists of the helix, antihelix, earlobe, and tragus. The mastoid process is positioned just behind the earlobe. The slightly curving auditory canal ends at the tympanic membrane, which transmits sound...

Immunostaining Phospho-epitopes in Ciliated Organs of Whole Mount Zebrafish Embryos

Techniques are described to immunostain phospho-epitopes in whole zebrafish embryos and then conduct two-color fluorescent confocal localization in cellular structures as small as primary cilia. The techniques for fixing and imaging can define the location and kinetics of the appearance or activation of specific proteins.

Auditory sensation, commonly called hearing, involves the transformation of sonic waves into neural impulses facilitated by the structures of the auditory organ. The prominent, flesh-like structure on the side of the head, called the auricle, directs sound waves towards the auditory canal. The auricle is often mislabeled as the pinna, a term more aligned with mobile structures like a feline's external ear. The auditory canal penetrates the cranium via the external auditory meatus of the...
