Semi-high Throughput

Semi-high throughput refers to an intermediate-scale experimental approach that processes more samples or conditions in parallel than conventional studies while retaining substantial experimental control and data quality. In developmental biology, it typically combines miniaturized assays, standardized culture conditions, parallel handling, and automated or semi-automated imaging to measure developmental phenotypes across multiple embryos, cells, or treatments. This balance between scale and flexibility supports systematic studies of gene function, signaling pathways, morphogenesis, and developmental toxicity without requiring the infrastructure of fully high-throughput screening. Semi-high-throughput workflows can accelerate hypothesis testing, reveal reproducible phenotypic patterns, and help prioritize conditions for detailed mechanistic investigation.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.

A nicotinamide adenine dinucleotide (NADH)-coupled ATPase assay has been adapted to semihigh throughput screening of small molecule myosin inhibitors. This kinetic assay is run in a 384-well microplate format with total reaction volumes of only 20 µL per well. The platform should be applicable to virtually any ADP producing enzyme.
