Dynamic Culture

Dynamic culture is the controlled growth of cells, tissues, or microorganisms under changing physical and chemical conditions rather than in a static environment. Engineering systems create these conditions through bioreactors, where fluid flow, mixing, perfusion, and regulated delivery of nutrients or oxygen influence mass transfer and cellular behavior. By reproducing mechanical cues such as shear stress while maintaining temperature, pH, and dissolved gases, dynamic culture can improve uniformity, viability, and tissue maturation. It supports applications in tissue engineering, regenerative medicine, bioprocess development, and biomaterials testing, while providing experimental models that better reflect the changing environments found in living systems.

Microfabricated Platforms for Mechanically Dynamic Cell Culture

In this protocol, we demonstrate the fabrication of a microactuator array of vertically displaced posts on which the technology is based, and how this base technology can be modified to conduct high-throughput mechanically dynamic cell culture in both two-dimensional and three-dimensional culture paradigms.

Fabrication of Micropatterned Hydrogels for Neural Culture Systems using Dynamic Mask Projection Photolithography

Simple techniques are described for the rapid production of microfabricated neural culture systems using a digital micromirror device for dynamic mask projection lithography on regular cell culture substrates. These culture systems may be more representative of natural biological architecture, and the techniques described could be adapted for numerous applications.

Use of pHluorin to Assess the Dynamics of Axon Guidance Receptors in Cell Culture and in the Chick Embryo

We describe here the use of a pH-sensitive green fluorescent protein variant, pHluorin, to study the spatio-temporal dynamics of axon guidance receptors trafficking at the cell surface. The pHluorin-tagged receptor is expressed both in cell culture and in vivo, using electroporation of the chick embryo.

Organotypic Slice Cultures to Study Oligodendrocyte Dynamics and Myelination

A technique to study NG2 cells and oligodendrocytes using a slice culture system of the forebrain and cerebellum is described. This method allows examination of the dynamics of proliferation and differentiation of cells within the oligodendrocyte lineage where the extracellular environment can be easily manipulated while maintaining tissue cytoarchitecture.
