Revolving Disc Test

The Revolving Disc Test is an engineering experiment that evaluates the mechanical behavior of a rotating disc under controlled operating conditions, helping relate rotational motion to measurable forces, torque, and resistance. During the test, the disc spins at a defined speed while applied loads, contact conditions, or attached components produce effects such as friction, centrifugal force, vibration, or deformation; instruments record the resulting response. Engineers use these measurements to assess rotational stability, energy loss, material performance, and wear, while comparing experimental results with theoretical models. The test supports the design, validation, and maintenance of rotating machinery, braking systems, drives, and other mechanical components.

The generation, purification and cell invasion of intracellular, cytoplasmic full length DISC1 protein aggresomes from cell cultures and of a labeled, multimeric recombinant DISC1 protein fragment in E. coli are described. Cell invasiveness is shown for recipient cells in cell culture and for neurons in vivo after stereotactical brain inoculation.

Understanding the neural substrates of behavior requires brain circuit ensemble recording. Because of its genetic tractability, the mouse offers a model for circuit dissection and disease mimicry. Here, a method of designing and fabricating miniaturized probes is described that is suitable for targeting deep brain structure in the mouse.

Source: Muto, A., et al. Ablation of a Neuronal Population Using a Two-photon Laser and Its Assessment Using Calcium Imaging and Behavioral Recording in Zebrafish Larvae. J. Vis. Exp. (2018) In this video, a two-photon laser ablation procedure is demonstrated, enabling the selective destruction of fluorescent neurons in agarose-immobilized zebrafish larvae. High-resolution imaging before and after the procedure confirms efficient neuronal damage, as observed by the absence of fluorescence.

Here, we present a protocol to use a curvelet transform-based, open-source MATLAB software tool for quantifying fibrillar collagen organization in the extracellular matrix of both normal and diseased tissues. This tool can be applied to images with collagen fibers or other types of line-like structures.
