Metabolite Derivatization

Metabolite derivatization is the chemical modification of small molecules to improve their detection, separation, or structural characterization, making it valuable for analyzing complex environmental samples. In this process, reactive functional groups such as hydroxyl, carboxyl, amino, or carbonyl groups are selectively converted into derivatives with altered polarity, volatility, ionization efficiency, or stability before chromatographic or mass spectrometric analysis. Environmental researchers use derivatization to enhance the measurement of metabolites in soil, water, sediments, and biological samples, including compounds present at trace concentrations. The approach can increase analytical sensitivity, resolve overlapping signals, and support more reliable assessments of biogeochemical processes, pollution, and ecosystem health.

Derivatization of Protein Crystals with I3C using Random Microseed Matrix Screening

This article presents a method to generate protein crystals derivatized with I3C (5-amino-2,4,6-triiodoisophthalic acid) using microseeding to generate new crystallization conditions in sparse matrix screens. The trays can be set up using liquid dispensing robots or by hand.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
