Dna Blotting

DNA blotting is a molecular biology technique used to identify specific DNA sequences within a complex mixture, making it valuable for genetic analysis. In a Southern blot, restriction enzymes cut genomic DNA, gel electrophoresis separates the fragments by size, and the DNA is denatured and transferred to a membrane; a labeled complementary probe then hybridizes with the target sequence for detection. This method can reveal restriction fragment length polymorphisms, gene rearrangements, insertions, deletions, and changes in copy number. Although newer amplification and sequencing methods are often faster, DNA blotting remains an important tool for validating genome structure and studying inheritance.

We present a method to quantify DNA methylation based on the 5-methylcytosine (5-mC) dot blot. We determined the 5-mC levels during chondrocyte dedifferentiation. This simple technique could be used to quickly determine the chondrocyte phenotype in ACI treatment.
