Library Complexity Analysis

Library complexity analysis is a quality-control method that estimates the number and diversity of unique DNA or RNA molecules represented in a sequencing library. It typically examines sequencing reads for duplicate sequences or uses unique molecular identifiers to distinguish original molecules from PCR copies, then models how additional sequencing would increase the recovery of new fragments. In genetics, this analysis helps identify overamplification, insufficient starting material, or preparation bottlenecks before downstream interpretation. It guides sequencing-depth decisions and strengthens the reliability of applications such as variant detection, gene-expression profiling, chromatin studies, and other next-generation sequencing experiments.

Biopanning bacterial display libraries is a proven technique for discovery of peptide affinity reagents, a robust alternative to antibodies. The semi-automated sorting method herein has streamlined biopanning to decrease the occurrence of false positives. Here we illustrate the thought process and techniques applied in evaluating candidates and minimizing downstream analysis.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.
