Nuclear Tau Function

Nuclear tau function refers to the activities of tau protein within the cell nucleus, extending beyond its well-known role in stabilizing neuronal microtubules. After entering the nucleus, tau can associate with DNA, chromatin, and nucleolar components, where its charge-dependent interactions may influence chromatin organization, gene regulation, and protection of genomic material from damage. In genetics, studying nuclear tau connects MAPT gene expression, alternative tau isoforms, and disease-associated mutations with changes in nuclear organization and neuronal survival. This research may clarify how abnormal tau localization contributes to neurodegenerative disease and identify molecular targets for investigating genome maintenance and neuronal dysfunction.

2012

A protocol for preparation of robust, small-scale HeLa nuclear extracts is described. This protocol is valuable for assays that require use of small populations of cells, such as cells treated with drugs or RNAi. The method should be applicable to a wide variety of gene expression assays and other cell types, including patient cells.

Unmodified and hyperphosphorylated tau proteins were used in two in vitro aggregation assays to reveal the hyperphosphorylation-dependent fast aggregation kinetics. These assays pave the way for future screens for compounds that can modulate the propensity of hyperphosphorylated tau to form fibrils that underlie the progression of Alzheimer’s disease.

2026

We describe a sensitive, time-resolved quantitative platform to measure clearance of extracellular tau in human induced pluripotent stem cell (iPSC)-derived microglia using a microplate-based luminescence system. This approach enables sensitive detection of tau degradation kinetics in a microwell format, allowing for high-throughput interrogation of tau handling under various experimental conditions.

This manuscript describes standard protocols for measuring tau hyperphosphorylation, measuring tau binding to microtubules, and localization of intracellular tau following drug treatments. These protocols can be used repetitively for screening drugs or other compounds that target tau hyperphosphorylation or microtubule binding.
