Cancer Cell Killing

Cancer cell killing is the destruction or elimination of malignant cells, a central goal of immune surveillance and cancer treatment. In immunology, cytotoxic T lymphocytes recognize tumor-associated antigens presented by major histocompatibility complex molecules, while natural killer cells detect abnormal patterns or reduced antigen presentation; both can release perforin and granzymes that trigger target-cell apoptosis. Researchers study these mechanisms to understand host defense, infection-related cancer risk, and immune escape, and to develop therapies such as adoptive cell transfer and immune checkpoint blockade. Measuring target-cell death also helps evaluate immune-cell potency and treatment response.

Many infections elicit a strong CTL response, but occasionally, the quantity of responding cells does not correlate to control of the pathogen1. One measure of CTL quality is their ability to kill specifically2. CFSE labeling of target cells can be used to investigate this CTL response quality in vivo3,4.

This protocol is designed to assess immunotherapeutic redirected T-cell (CAR T-cell) cytotoxicity against 3D structured cancerous cells (spheroids) in real time.

Assessment of Opsonophagocytic Killing Activity against Bacterial Pathogens

The video shows an in vitro opsonophagocytic killing assay using a co-culture of human neutrophils with Streptococcus pneumoniae. Antibodies and complement proteins facilitate opsonization of the bacteria, enabling neutrophils to recognize and kill bacteria, showing successful neutrophil opsonophagocytic activity.
