Cell Lysate Purification

Cell lysate purification is the process of separating proteins, nucleic acids, organelles, and other biomolecules from a disrupted cell mixture to obtain a cleaner sample for analysis. It typically combines cell lysis with centrifugation, filtration, precipitation, or chromatography, using differences in size, density, solubility, charge, or affinity to isolate target components while removing contaminants. In immunology and infection research, purified lysates support antigen detection, antibody characterization, enzyme assays, and studies of host-pathogen interactions. Careful control of lysis conditions, temperature, protease activity, and buffer composition helps preserve molecular structure and improves the reliability of downstream experiments.

We describe here a novel, robust, and efficient tandem affinity purification (TAP) method for the expression, isolation, and characterization of protein complexes from eukaryotic cells. This protocol could be utilized for the biochemical characterization of discrete complexes as well as the identification of novel interactors and post-translational modifications that regulate their function.
