Degranulation Assay

A degranulation assay is a laboratory test that measures the release of preformed secretory granules from immune cells, providing a functional readout of cellular activation. When receptors recognize a target or stimulus, signaling raises intracellular calcium and drives granule fusion with the plasma membrane, exposing markers such as CD107a and releasing mediators including granzymes, perforin, or histamine. Researchers use flow cytometry, microscopy, or mediator-specific measurements to quantify responses in natural killer cells, cytotoxic T cells, mast cells, and other leukocytes. In immunology and infection research, the assay helps evaluate cytotoxic function, pathogen-induced activation, immune deficiencies, and responses to vaccines or therapies.

This video demonstrates a colorimetric assay to determine the degranulation activity of mast cells. The enzyme released by the mast cells hydrolyzes the substrate and produces a colored product. The degranulation activity of the mast cells was measured as the percentage release of enzymes outside the cell normalized with the total enzyme content in the cell.
