Endosomal Transport

Endosomal transport is the cellular system that sorts and moves internalized molecules, membrane proteins, and pathogens through membrane-bound compartments, helping maintain cell organization and regulate signaling. After endocytosis, cargo enters early endosomes, where Rab GTPases and tethering factors coordinate vesicle formation, fusion, and sorting toward recycling endosomes, late endosomes, or lysosomes; SNARE proteins help drive membrane fusion. In immunology, this trafficking controls antigen processing, receptor recycling, and the delivery of immune signaling components. Infectious agents can exploit or disrupt these pathways to enter cells, avoid degradation, or alter host defenses, making endosomal transport important for understanding disease mechanisms and therapeutic targets.

In Vitro Polymerization of F-actin on Early Endosomes

Early endosome functions depend on F-actin polymerization. Here, we describe a microscopy-based in vitro assay that reconstitutes the nucleation and polymerization of F-actin on early endosomal membranes in test tubes, thus rendering this complex series of reactions amenable to biochemical and genetic manipulations.
