Ex Vivo Brain Slices

Ex vivo brain slices are thin sections of brain tissue maintained outside the organism in a controlled culture environment, preserving local cellular architecture and interactions for experimental study. Typically prepared from freshly isolated brain and supported in oxygenated artificial cerebrospinal fluid, the slices remain viable while researchers can apply pathogens, immune stimuli, or drugs and monitor responses through microscopy, electrophysiology, and molecular assays. In immunology and infection research, this model enables analysis of microglia, astrocytes, neurons, and immune-cell responses during neuroinflammation, pathogen invasion, and tissue injury. It bridges simplified cell cultures and whole-animal studies while allowing precise, localized manipulation of living neural tissue.

Optogenetic approaches are widely used to manipulate neural activity and assess the consequences for brain function. Here, a technique is outlined that upon in vivo expression of the optical activator Channelrhodopsin, allows for ex vivo analysis of synaptic properties of specific long range and local neural connections in fear-related circuits.

Whole-cell Patch-clamp Recordings in Brain Slices

This protocol describes basic procedural steps for performing whole-cell patch-clamp recordings. This technique allows the study of the electrical behavior of neurons, and when performed in brain slices, allows the assessment of various neuronal functions from neurons that are still integrated in relatively well preserved brain circuits.

In vivo Postnatal Electroporation and Time-lapse Imaging of Neuroblast Migration in Mouse Acute Brain Slices

Neuroblast migration is a fundamental event in postnatal neurogenesis. We describe a protocol for efficient labeling of neuroblasts by in vivo postnatal electroporation and subsequent visualization of their migration using time-lapse imaging of acute brain slices. We include a description for the quantitative analysis of neuroblast dynamics by video tracking.

Neuronal membrane trafficking dynamically controls plasma membrane protein availability and significantly impacts neurotransmission. To date, it has been challenging to measure neuronal endocytic trafficking in adult neurons. Here, we describe a highly effective, quantitative method to measure rapid changes in surface protein expression ex vivo in acute brain slices.
