Water Oil Particles

Water-oil particles are dispersed structures in which aqueous droplets containing biological materials are suspended within an oil phase, creating compartments that can protect and deliver their contents. Their properties depend on emulsification, interfacial stabilization, droplet size, and the controlled release of dissolved antigens or other immune-active molecules. In immunology and infection research, water-oil particles support vaccine formulation by presenting antigens with adjuvant activity, enhancing immune stimulation, and regulating exposure to host immune cells. Studying their composition and behavior helps researchers improve antigen delivery, evaluate vaccine responses, and design more effective strategies against infectious diseases.

This manuscript describes a simple and high-throughput method for assembling water-soluble proteins with hydrophobic pigments that is based on water-in-oil emulsions. We demonstrate the effectiveness of the method in the assembly of native chlorophylls with four variants of recombinant water-soluble-chlorophyll binding proteins (WSCPs) of Brassica plants expressed in E. coli.

The low-energy water-accommodated fraction (LEWAF) of crude oil is a challenging system to analyze, because over time, this complex mixture undergoes chemical transformations. This protocol illustrates methods for the preparation of the LEWAF sample and for performing photo-irradiation and chemical analysis by trapped ion mobility spectrometry–FT-ICR MS.

We illustrate the methods involved in screening and identification of the biosurfactant producing microbes. Methods for chromatographic characterization and chemical identification of the biosurfactants, determining the industrial applicability of the biosurfactant in enhancing residual oil recovery are also presented.
