Granulocyte Recruitment

Granulocyte recruitment is the coordinated movement of neutrophils, eosinophils, and basophils from the bloodstream into tissues where infection or injury occurs, making it a central process in innate and inflammatory immunity. In response to microbial signals or tissue damage, endothelial cells and resident immune cells release chemokines and other mediators that promote selectin-dependent rolling, integrin-mediated firm adhesion, and diapedesis, after which granulocytes follow chemical gradients toward the affected site. Studying this process helps explain how infections are contained, how inflammation is regulated, and why excessive or misdirected recruitment can damage tissues. Experimental models of granulocyte trafficking also support research into inflammatory disease and potential therapies.

The ability of inflamed endothelium to recruit leukocytes from flow is regulated by mesenchymal stromal cells. We describe two in vitro models incorporating primary human cells that can be used to assess neutrophil recruitment from flow and examine the role that mesenchymal stromal cells play in regulating this process.
