Hemagglutinin Glycoprotein

Hemagglutinin glycoprotein is a viral surface protein, best known for enabling influenza viruses to attach to and enter host cells, making it central to infection and immune recognition. It binds sialic acid receptors on respiratory epithelial cells, is taken into endosomes, and undergoes an acid-triggered structural change that drives fusion between the viral envelope and cell membrane. Antibodies targeting hemagglutinin can block receptor binding or membrane fusion, providing a major basis for protective immunity and influenza vaccine design. Studying its antigenic variation also supports surveillance, diagnostic development, and the evaluation of broadly protective vaccines and antiviral strategies.

Here we describe a way to express correctly folded and functional influenza virus surface antigens derived from the novel Chinese H7N9 virus in insect cells. The technique can be adapted to express ectodomains of any viral or cellular surface proteins.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.
