Hiv-specific Cd4 T Cells

HIV-specific CD4 T cells are helper T lymphocytes that recognize HIV-derived peptides and coordinate immune responses against infection. Antigen-presenting cells display these peptides on major histocompatibility complex class II molecules, allowing T-cell receptors to trigger activation, cytokine production, and support for B cells and HIV-specific CD8 T cells. Because HIV can infect CD4 T cells through the CD4 receptor and chemokine coreceptors, these virus-responsive cells may also become targets for viral replication, depletion, or latent infection. Studying their frequency, function, and phenotype supports immune monitoring, vaccine evaluation, and research into viral persistence and strategies for durable HIV control.

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.

Cytotoxicity assays to measure natural killer cell lytic responses to HIV-infected cells is limited by the purity of the target cells. We demonstrate here the isolation of a highly purified population of HIV-1 infected primary T-cell blasts by taking advantage of HIV-1 s ability to down-modulate CD4.

Several 2’-Fluoro RNA aptamers against HIV-1Ba-L gp120 with nanomole affinity are isolated from a RNA library by in vitro SELEX procedure. A new dual inhibitory function anti-gp120 aptamer-siRNA chimera is created and shows considerable promise for systemic anti-HIV therapy.
