Light-sheet Microscopy

Light-sheet microscopy is an optical imaging technique that illuminates a specimen with a thin sheet of light while collecting fluorescence from the side, enabling rapid three-dimensional visualization with reduced photobleaching and phototoxicity. By restricting excitation to a single optical plane and scanning through the sample, it produces high-resolution volumetric datasets while preserving temporal information in living or cleared tissues. In immunology and infection research, the method can track immune-cell migration, host-pathogen interactions, and the spatial organization of infected tissues or organoids. These capabilities support quantitative analysis of cellular behavior and disease progression across scales that conventional microscopy may not capture.

Light sheet fluorescence microscopy is an excellent tool for imaging embryonic development. It allows recording of long time-lapse movies of live embryos in near physiological conditions. We demonstrate its application for imaging zebrafish eye development across wide spatio-temporal scales and present a pipeline for fusion and deconvolution of multiview datasets.

This protocol shows a plant sample preparation method for light-sheet microscopy. The setup is characterized by mounting the plant vertically on the surface of a gel and letting it grow in controlled bright conditions. This allows long-term observation of plant organ development in standardized conditions.

Lattice Light Sheet Microscopy to Visualize Receptor-Ligand Interactions in Live Cells

In this video, we describe the lattice light-sheet microscopy technique to visualize interactions between live T-cells and antigen-presenting cells. The method involves focusing an ultrathin lattice light sheet on an interacting cell pair to obtain a series of two-dimensional images, which are combined to obtain high-resolution four-dimensional images that reveal the spatiotemporal dynamics of the cell-cell interaction.
