Mouse Colon Isolation

Mouse colon isolation is the controlled removal of the colon from a mouse for anatomical, cellular, or molecular analysis, making it a foundational procedure in gastrointestinal immunology and infection research. After euthanasia, researchers expose the abdominal cavity, separate the colon from surrounding tissues, and preserve or process the tissue for downstream assays such as histology, cell isolation, RNA analysis, or pathogen measurement. This approach enables direct assessment of epithelial integrity, mucosal immune responses, inflammatory lesions, and microbial effects. By linking tissue-level changes to infection or immune activation, colon isolation supports studies of host-pathogen interactions, intestinal disease mechanisms, and potential therapeutic interventions.

2013

The myofibroblast is an influential stromal cell of the gastrointestinal tract that regulates important physiologic processes in both normal and disease states. We describe a technique that allows for the isolation of primary myofibroblasts from both mouse and human colon tissue, which can be utilized for in vitro experimentation.

In vitro Organoid Culture of Primary Mouse Colon Tumors

A simple method to establish primary murine colon tumor organoid is described. This method utilizes the feature that colon tumor cells survive and grow into organoids in media containing limited growth factors, whereas normal colon epithelial do not.

A progressive colonization procedure is described to further assess its impact on the host hepatic metabolism. Colonization is monitored non invasively by evaluating the urinary excretion of microbial co-metabolites by NMR-based metabolic profiling while hepatic metabolism is assessed by High Resolution Magic Angle Spinning (HR MAS) NMR profiling of intact biopsy.

(2019)This video demonstrates a method for preparing infected mouse kidney tissue, staining DNA, and detecting bacterial gene expression using dual fluorescent reporters. Red marks all bacteria, and green highlights infection-responsive cells.

Electrophysiological Recording from Colonic Afferent Nerves in an Ex Vivo Rat Colon
