Radiolabeled Nucleotides

Radiolabeled nucleotides are nucleotides tagged with radioactive isotopes, enabling researchers to track nucleic acid synthesis, metabolism, and molecular interactions through emitted radiation. Cells or biochemical systems incorporate these labeled building blocks into DNA or RNA, and instruments detect the resulting radioactive signal to measure the location, amount, or timing of specific processes. In immunology and infection research, radiolabeled nucleotides help quantify pathogen replication, monitor lymphocyte proliferation, analyze immune-cell responses, and study nucleic acid-based assays. Their measurable signals provide sensitive experimental readouts for investigating host-pathogen interactions and evaluating changes caused by antimicrobial or immunomodulatory treatments.

This paper describes a method for the measurement of fuel oxidation in Drosophila melanogaster in which trace amounts of specific radiolabeled metabolic substrates are fed to flies. The exhaled radiolabeled CO2 that is a produced from fuel oxidation is collected and measured.

Protein kinases are highly evolved signaling enzymes and scaffolds that are critical for inter- and intracellular signal transduction. We present a protocol for measuring kinase activity through the use of radiolabeled adenosine triphosphate ([γ-32P] ATP), a reliable method to aid in elucidation of cellular signaling regulation.
